Rune, ranked No 16, will face the winner of the match between Canadian Felix Auger-Aliassime and Jack Draper next.
Ahead of his victory, the 21-year-old Dane edged out Italian wildcard Matteo Berrettini (2-6, 6-1, 6-4) and won against Portuguese qualifier Nuno Borges (6-3, 7-6 (8)).
Monfils, ranked No 46, defeated Australian Alexei Popyrin (7-5, 6-3) and edged out Spaniard Carlos Alcaraz, the second seed (4-6, 7-6 (5), 6-4) earlier in the tournament.
